Promoter hypermethylation of IRX3 causes reduction of gene expression in vitro. GC content summary of IRX3 promoter region with the locations for the pCpGL cloning primers and MeDIP primers. Plasmid map of pCpGL-IRX3 (5216 bp) with IRX3 promoter sequence (1364 bp) inserted before the firefly luciferase section. Quantification of normalized luciferase activity showed a 143.4-fold reduction for the methylated construct as compared to the unmethylated control. Red color indicates a high amount of GC content, whereas blue color represents low GC content. One-way ANOVA was used for multiple group comparison.
