Inhibition of DOT1L leads to increased sensitivity to DNA damaging agents. a SW837 cells were pretreated with either with DMSO or the DOT1L inhibitor EPZ5676 (1 μM) for 3 h followed by treatment with NCS (100 ng/ml) for the indicated time points. Whole cell lysates were immunoblotted and detected with the indicated antibodies. b Cell proliferation assay with SW837 cells treated either with 5-fluorouracil (5-FU; 5 μM), Irinotecan (IRI; 1 μM), EPZ5676 (1 μM) individually or in combination as indicated.
