Epigenetic silencing mechanism of PcG proteins. Following the recruitment of PRC2 to chromatin, the histone methyltransferase E(Z) catalyzes trimethylation onto H3K27. Subsequent recruitment of PRC1 occurs through affinity binding of the chromodomain of PC subunit to H3K27me3. The PRC1 dRING monoubiquitylates onto H2K119 which further consolidates the transcriptional repression and enhances the chromatin compaction. Pho-RC comprises of PHO and SFMBT which binds to PRE via DNA binding activity of PHO. PHO then recruits PRC2 which methylate local chromatin
